FISH using X and/or Y chromosome probs helps detect abnormalities in the copy number of chromosomes X and Y on lymphocites, uncultured amniotic fluid or buccal mucosa cells.
Specimen requirement -
- for prenatal screen 20 to 30 ml amniotic fluid collected in 2 to 3 sterile tubes. The first few mls of fluid should be discarded as they increase the likelihood of maternal cell contamination. Specimen should be kept at room temperature; do not freeze or refrigerate. Specimen should be sent at the laboratory immediately. The following information should accompany the specimen: patient's name, physician's name, date collected, EGA by ultrasound, LMP (if available), maternal age, and indication for study.
- for mosaicism studies 2 ml of peripheral blood collected in an heparin (green top) vacutainer tube or oral smears obtained by scraping the inner cheek (contact the specialists)
Reporting time - FISH results for prenatal screen are usually available within 2-3 days. FISH results for peripheral blood and buccal mucosa screen are usually available within 7-10 days.
